Spheroplasts were generated from Borrelia burgdorferi by application of Tris-buffered lysozyme and EDTA. Cultures were examined during preparation by scanning electron microscopy (SEM) and by negative staining and/or thin sectioning followed by transmission electron microscopy (TEM). Spirochete and spheroplast proteins were also compared by electrophoresis to determine the effect of outer membrane manipulation during spheroplast preparation on the amounts of two outer surface proteins, OspA and OspB. Periplasmic flagellar release from the confines of the outer membrane began at the earliest steps of the preparation process and increased with continued progress toward the spheroplast condition. With the addition of Tris alone, spirochete uncoiling and outer membrane disruption became apparent. Lysozyme addition resulted in the formation of electron-transparent blebs where the outer membrane separated from the cytoplasmic membrane. Despite the permeability of the outer membrane to the lysozyme and the apparent alterations in its structure, OspA and B showed no decrease in concentration throughout the spheroplast preparation process. It is suggested that the flagellar release that occurs during spheroplast production also occurs in the host during Lyme disease, accounting for early immune responses to flagellin.

Adhesion to and invasion of cultured tick (Acarina: Ixodidae) cells by Borrelia burgdorferi (Spirochaetales: Spirochaetaceae) and maintenance of infectivity.

Lyme disease spirochetes, Borrelia burgdorferi, interact with cultured tick cells in ways similar to those reported to occur in the vector Ixodes dammini Spielman, Clifford, Piesman & Corwin. Spirochete adhesion and penetration were examined using a cell line from embryos of Rhipicephalus appendiculatus Neumann that morphologically resembles tick gut cells, RAE25. Cocultivation of B. burgdorferi with these cells permitted prolonged maintenance of infectivity for hamsters. Borrelial adherence to RAE25 cells was time- and density-dependent and increased by 10-15% per h during the first 5.5 h of cocultivation when we used a concentration of 4 x 10(7) spirochetes/ml. After 6 h, > 90% of the cells bound an average of 3-5 spirochetes per cell. Low passage, hamster-infective strains of B. burgdorferi (JMNT and CD16) showed a 2-3-fold higher rate of adhesion to RAE25 cells than the highly passaged, noninfectious strain B31. Inactivation of CD16 or JMNT by heat, starvation, or treatment with puromycin reduced adherence by 40-60%, whereas pretreatment with monoclonal antibodies to the outer surface proteins had no effect. Spirochetes adhered to young I. dammini cell lines to a similar degree as they did to RAE25, whereas lines from the ticks Dermacentor variabilis (Say) (RML15) and Boophilus microplus (Canestrini) (BME26) bound 30-60% fewer spirochetes. Electron microscopy revealed epicellular borreliae associated with coated pits and vesicles before endocytosis, and intracellular spirochetes were surrounded by a host cell-derived membrane.

Nevertheless, she was re-treated with 2 weeks of parenteral ceftriaxone in 1989 that had no effect on her neurologic symptoms. During the time of observation, this patient died. At autopsy, lymphoid mononuclear cells were observed surrounding the intracerebral vessels in one section. Using Dieterle silver stain, a spirochete was present in the cortex and another was exterior to a leptomeningeal vessel."

Either NYMC is taking these scientists for fools, or their is some formal cooperation to publish fraudulent science over the persistence of spirochetes, especially as dormant forms, and as regards intracellularity, both of which mean these spirochetes are not all killed by antibiotics, and that, Lyme is an incurable relapsing fever borreliosis (which we of course already knew).  Anyone who has Chronic Lyme knows this is a real chronic relapsing illness.  It's not something we all decided to imaginate together.
